Shafi Kamalbasha,Manuel J. A. Eugsterconjugated alkaloids in 24-hour urine samples from tobacco users are 1.0, 0.5 and 2.3 for nicotine, cotinine and .-3′-hydroxycotinine, respectively. Nicotine, cotinine and .-3′-hydroxycotinine and their glucuronides together with the N-oxides of nicotine and cotinine, account for more than 90% of the ingested dose of nicotine.

Oxidative Metabolism of Nicotine ,ute of metabolism in most mammalian species involves oxidation of the 5′-carbon atom to give the γ-lactam derivative cotinine. Oxidation of the N-methyl group results in demethylation to nornicotine, which is a minor metabolite in humans. Nicotine also undergoes oxidation of the pyrrolidine nitrogen

Detection of Nornicotine in Human Urine after Infusion of Nicotinernicotine has been found in the urine presenting evidence for the existence of this metabolic step. But, only less than 1% of the administered dose of nicotine is excreted as nornicotine possibly indicating rapid further metabolism.
