描述A cutting-edge collection of readily reproducible molecular techniques to better understand, classify, and treat lymphoma. Among the highlights are methods to use immunoglobulin gene rearrangements as markers of clonality, to exploit patterns of somatic mutation in the variable regions to indicate at which stage transformation occurred, and to apply gene arrays to the question of biological heterogeneity in morphologically similar diseases. Research methodologies that are highly likely to become routine practice in the future, such as DNA microarray and immunoglobulin V-gene rearrangements, and measurement of minimal disease, are included. There are also molecular techniques for providing for producing novel therapeutics, such as a DNA vaccine with patient-specific sequences derived from the lymphoma in question.

Follicular Lymphoma,s not yet possible to determine whether the detection of residual lymphoma cells by PCR is an indication to continue therapy. Therefore, although these techniques provide a useful adjunct to standard methods of detection and diagnosis, their role in staging of disease outcome remains investigational.

Molecular Diagnosis of Lymphoma,and outcome using microarray technology were generated using DLBCL samples and are described. The different types of microarray platform and data analysis tools are reviewed followed by a detailed step-by-step guide to data generation using the Affymetrix chip system from RNA extraction to laser scanning of the hybridized and stained chips.

Antibody Techniques Used in the Study of Anaplastic Lymphoma Kinase-Positive ALCL,cal category of anaplastic large cell lymphoma. Antibody techniques have also provided major new insights into the biology of these tumors. This chapter describes the use of antibodies in both immunolabeling (immunocytochemical and immunofluorescence) procedures and in biochemical procedures in the study of ALK-positive ALCL.

We also describe how to prepare this material for fluorescence . hybridization techniques, both for further delineation of complex chromosomal rearrangements and for interphase analysis in cases in which conventional cytogenetics has not been successful.
