描述A characteristic property of most, or perhaps all, proteins is their ability to combine specifically and reversibly with various substances. Well known examples are enzymes that bind substrates and inhibitors, and antibodies that bind antigens. This book deals with lectins, a class of proteins that bind carbohydrates. Another characteristic property of lectins is that they agglutinate cells or precipitate polysaccharides and glycoproteins. This is because lectins are polyvalent, i.e. each lectin molecule has at least two carbohydrate binding sites to allow crosslinking between cells (by combining with sugars on their surfaces) or between sugar containing macromolecules. The agglutinating and precipitating activities of lectins are very similar to those of antibodies. They can likewise be specifically inhibited by low molecular weight compounds (haptens), which in the case of lectins are sugars or sugar containing compounds (Fig. 1.1). Functions in nature,lly regulated and that their appearance often coincides with a distinct physiological change in the life of an organism. Notably, the putative receptors for the lectins, i.e. complementary carbohydrate structures, are frequently also under strict developmental control.

Epilogue,oanatomical pathways and bone marrow transplantation. There is little doubt that as we learn more about lectins, and discover new ones, novel and unexpected applications can be anticipated. We may also look forward to a time when lectins with desired specificities will be designed and produced by recombinant DNA techniques.

Carbohydrate specificity,ins is relatively weak, does not result in the formation of covalent bonds, and is reversible, like the reaction of an antibody with an antigen (or of an enzyme with an inhibitor). The compound that inhibits at the lowest concentration is assumed to be most complementary to the combining site of the lectin.

Biological activities,d for the development of new ones; they provide us with a means to investigate different cellular and subcellular processes, especially those initiated at the cell surface; and last, but not least, the biological activities of lectins detected in the laboratory give clues to the probable functions of these substances in nature.

Applications, soluble substances and cells can be reversed by simple sugars. The specific interaction of a biopolymer with a lectin may be taken as evidence that the polymer contains carbohydrate. Thus, lectin binding has been widely used to demonstrate that membrane receptors for many hormones, growth factors, neurotransmitters and toxins are glycoconjugates.
