Assessment of RET/PTC Oncogene Activation in Thyroid Nodules Utilizing Laser Microdissection Followedules that exhibit borderline morphological signs of papillary cancer represent a diagnostic challenge. Rearrangements of the RET oncogene have been proposed as a marker for papillary thyroid cancer. In this chapter, methods for the analysis of the RET oncogene in laser microdissected papillary thyroid cancer tissue are described.

Methacarn Fixation for Genomic DNA Analysis in Microdissected Cellsmmunostaining, fragments of about 180 bp can be amplified with 150-270 cells by single-step PCR. These features demonstrate the suitability of methacarn-fixed wax-embedded tissue for practical genomic DNA analysis in terms of tissue handling, extraction efficiency, and satisfactory PCR results.

Laser-Assisted Microdissection of Membrane-Mounted Sections Following Immunohistochemistry and In Sitimized protocols that allow IHC to be performed for detecting a wide range of antigens in conjunction with LMM, both on formalin-fixed paraffin-embedded and on frozen sections. In addition, we present a quick, versatile protocol for performing ISH on archival material suitable for LMM.

Laser-Assisted Cell Microdissection Using the PALM Systemp53 mutations from single cells is given. The described procedures emphasize preparation of tissue, laser microdissection including catapulting of cells, and extraction of RNA and DNA. Downstream experiments for validation are also shown.
