The Surface Layers of ,, division alternating at right angles to the previous division to form square arrays (Murray 1963, Kuhn and Starr 1965). Cells within the sheet are held together by slender fibres, which are disposed between adjacent cells, as well as between the outer membrane and RS-lavers (Chapman et al. 1963).

Three-Dimensional Structure of Surface Layers from Various , and , Species,c value (Baumeister et al. 1986). However, 3-D reconstructions are required (besides primary structure information) to describe and to compare the S-layers in greater detail. Here, the 3-D structures of S-layers of various . and . species are compared that are phylogenetically distant on the species, the genus, and the family level (Fig.1).

Achim Schmid,Pascal Siemsen,Ralf G?tzefor a first survey was freezeetching, more specifically deep-etching, in conjunction with surface relief reconstruction (Prüschenk et al. 1987). This technique has the merit to provide, albeit at relatively low resolution (2.5 to 3 nm), three-dimensional information about structures in situ (Baumeister et al. 1986).
