While many factors contribute to a high failure rate, it is somewhat surprising that poor pharmacokinetic properties, such as poor oral bioavailability, account for almost half of the failures in clinical development.

Allenberges and the width of the therapeutic window between effective and toxic blood concentrations, specific subpopulations might require a dose regimen which diverges from the population average regimen.
