Preparation of Intrinsically-Labelled Kininsand 5% Rh/CaCO. in H.O and DMF (1:1) plus 10 Ci of .H.. [4-.H-Phe.]-BK was obtained at 6.7 Ci/mmole in an overall yield of 15%. [4-.HPhe.]-BK was prepared similarly to yield an intrinsically-labelled peptide with a specific radioactivity of 21 Ci/mmole.

A Simple Radioassay for Human Urinary Kallikreins obtained on molecular sieve chromatography (Sephacryl), only one urinary enzyme possesses the ability to hydrolyze our substrate. The enzyme MW 45,000, is inhibited by Trasylol but not by soya bean trypsin inhibitor (SBTI). It is reactive with and is inhibited by antibodies prepared against pure HUK.

Assay Methods for Prekallikrein and Kininogens and Their Applicationsduction of prekallikrein and HMW-kininogen, not of LMW-kininogen, was found in pleural fluid of rat carrageenin pleurisy, and in plasma after i.v. injection of bromelain in rats. Determination of Prekallikrein in Plasma by Means of a Chromogenic Tripeptide Substrate for Plasma Ktial for the activation of prekallikrein in our method. Further experiments showed that the method was fairly selective and was not influenced by inhibitors present in normal plasma. The later finding was attributed to the high dilution of plasma made possible by using a potent activator and a sensitive substrate.
